Biological Background: CYP4F12 Function and Localization
- CYP4F12 encodes a cytochrome P450 monooxygenase of the CYP4 family, which primarily metabolizes fatty acids and xenobiotics.
- Catalyzes the hydroxylation of arachidonic acid to 18-hydroxy arachidonate with preference for the omega-2 position (PubMed:11162607). Related references: PMID:11162607
- Mediates epoxidation of double bonds in polyunsaturated fatty acids such as docosapentaenoic and docosahexaenoic acids (PubMed:16112640). Related references: PMID:16112640
- Shows low omega-hydroxylase activity toward leukotriene B4 and arachidonate (PubMed:11162645). Related references: PMID:11162645
- Involved in xenobiotic metabolism; hydroxylates the antihistamine drug ebastine (PubMed:11162645). Related references: PMID:11162645
- Localizes to the endoplasmic reticulum and microsomal membranes.
- Expressed in the small intestine, liver, colon, and heart.
- The protein contains a heme iron cofactor, features a transmembrane helix, and undergoes alternative splicing (UniProt keywords).
Experimental Guidance and Technical Tips
- The antibody is derived from rabbits immunized with a recombinant protein covering amino acids 265–524 of human CYP4F12. This region lies in the C-terminal half of the protein, which may contribute to specificity.
- Recommended for Western blotting (WB) and ELISA applications. Optimize dilution factors empirically using positive control lysates from liver or small intestine, which express CYP4F12.
- Predicted molecular weight is approximately 60 kDa; confirm using appropriate molecular weight markers.
- Cross-reactivity is reported for human, mouse, and rat samples. Validate detection in each species individually with appropriate controls.
- As a polyclonal antibody, lot-to-lot consistency should be verified for critical experiments. Consider using the same lot for longitudinal studies.
- For ELISA, pair with a suitable detection antibody; assess potential cross-reactivity with related CYP4F isoforms.
