Biological Background: CYP46A1 Function and Localization
- CYP46A1 (Cholesterol 24-hydroxylase, also known as CP46, CYP46) is a member of the cytochrome P450 superfamily, functioning as a heme-thiolate monooxygenase.
- It primarily catalyzes the 24S-hydroxylation of cholesterol in the brain, a key step for cholesterol elimination from neurons and overall brain cholesterol homeostasis (PubMed:10377398, PubMed:14640697, PubMed:18621681). Related references: PMID:10377398 PMID:14640697 PMID:18621681

- The enzyme is localized to the endoplasmic reticulum membrane, microsomes, and also postsynaptic and presynaptic regions of neurons, including dendritic projections.

- The immunogen is a synthetic peptide corresponding to residues 400–500 of human CYP46A1. This region may be surface-exposed; consider potential interference from protein–protein interactions or post-translational modifications near this epitope when interpreting results.
- For Western blot (WB), the predicted molecular weight is 57 kDa. Validate band specificity by comparing with a negative control or using a blocking peptide if available.
- For immunofluorescence on paraffin-embedded sections (IF-P), optimize antigen retrieval, blocking, and antibody dilution according to your sample type. A nuclear counterstain is recommended to differentiate cytoplasmic/membrane staining.
