Biological Background: Cytochrome P450 3A4 Function and Localization
- CYP3A4 is a major cytochrome P450 monooxygenase involved in the oxidative metabolism of a wide range of endogenous substrates including sterols, steroid hormones, retinoids, and fatty acids. Related references: PMID:10681376, PMID:11093772, PMID:11555828
- As a monooxygenase, it catalyzes the hydroxylation of carbon-hydrogen bonds using NADPH via cytochrome P450 reductase, playing a pivotal role in hormone biosynthesis and degradation, such as the formation of hydroxyestrogens from estrogens and the oxidative deactivation of testosterone. Related references: PMID:11555828, PMID:12865317, PMID:14559847, PMID:15373842, PMID:15764715, PMID:22773874, PMID:2732228
- CYP3A4 contributes to cholesterol metabolism by catalyzing hydroxylation at the C-4β and C-25 positions, generating oxysterols that may participate in cholesterol degradation and bile acid synthesis. Related references: PMID:21576599
- It is also responsible for the metabolism of xenobiotics, including many clinically administered drugs, by catalyzing reactions such as sulfoxidation of albendazole, hydroxylation of quinine, and monooxygenation of 1,8-cineole. Related references: PMID:11159812, PMID:10759686, PMID:8968357, PMID:11695850
- CYP3A4 localizes primarily to the membrane of the endoplasmic reticulum and microsomes.
- Tissue-specific expression is highest in liver and prostate, with detectable levels in intestine, kidney, adrenal gland, and other tissues, but variable expression reported in brain. Related references: PMID:19094056, PMID:18545703, PMID:19359404
- The protein is classified under keywords including Endoplasmic reticulum, Heme, Iron, Monooxygenase, Oxidoreductase, Lipid metabolism, and Steroid metabolism.
Experimental Guidance and Technical Tips
- The immunogen peptide (amino acids 400-503) resides in the C-terminal region of CYP3A4; researchers may consider this epitope for specific detection of the full-length protein.
- For Western blot (WB), a band at approximately 57 kDa is expected; optimize blocking and antibody dilution based on your sample type.
- For immunofluorescence (IF-P), ensure appropriate permeabilization and fixation; validate the staining in CYP3A4-expressing cell lines.
- For ELISA, the antibody may be employed as a capture or detection reagent; pair with a suitable secondary antibody and validate in the relevant sample matrix.
