Biological Background: CYP27A1 Function and Localization
- Cytochrome P450 27A1 (CYP27A1), also known as sterol 26-hydroxylase or vitamin D3 25-hydroxylase, is a mitochondrial monooxygenase essential for cholesterol metabolism and bile acid synthesis.
- Catalyzes the regio- and stereospecific hydroxylation of cholesterol and its derivatives, converting the terminal methyl group to a carboxylate in a three-step reaction; this step is critical in both the classic and alternative bile acid pathways. Related references:
- Generates active oxysterol ligands, such as 27-hydroxycholesterol, that act on NR1H2 and NR1H3 nuclear receptors to regulate lipid metabolism gene transcription. Related references:
- Hydroxylates 7-ketocholesterol to form 7-keto,27-hydroxycholesterol, which can activate the smoothened (SMO) signaling pathway (By similarity).
- Mediates cholestanol hydroxylation in the cerebellum, facilitating sterol diffusion through the blood-brain barrier and systemic clearance. Related references:
- Contributes to vitamin D3 activation via 25-hydroxylation, a redox step required for full hormonal activity. Related references:
- Localized to the mitochondrial inner membrane, consistent with its role in sterol processing.
- Expressed in neural retina, retinal pigment epithelium, and cerebellar gray/white matter at the protein level. Related references:
- Mutations in CYP27A1 cause cerebrotendinous xanthomatosis (CTX), a rare lipid storage disease affecting bile acid synthesis.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 200–400) covers a portion of the catalytic domain; ensure the antibody epitope is accessible under your experimental conditions.
- For Western blot, a band of approximately 60 kDa is expected; consider using mitochondrial-enriched lysates or whole-cell extracts from liver or retina as positive controls.
- Validate antibody specificity by comparing signal in CYP27A1‑overexpressing cells versus knockdown/knockout models.
- For ELISA, titrate the antibody using recombinant CYP27A1 or peptide antigen to determine the optimal working dilution.
- Cross‑reactivity with mouse and rat enables translational studies; however, verify cross‑reactivity in your specific tissue or cell line.
