Biological Background: CYP26A1 Function and Localization

  • CYP26A1 is a cytochrome P450 monooxygenase that belongs to the CYP26 family; it is also known as cytochrome P450RAI or P450RAI1, and plays a critical role in retinoic acid (RA) catabolism.
  • This enzyme catalyzes the hydroxylation of all-trans-retinoic acid (atRA) primarily at the C-4 and C-18 positions, initiating the degradation pathway that limits atRA biological activity and controls its homeostasis and signaling.

  • CYP26A1 is also capable of metabolizing other retinoic acid isomers, including 9-cis-RA, 13-cis-RA, and 9,13-di-cis-RA, and can oxidize all-trans-13,14-dihydroretinoate (DRA) to various metabolites.
