Biological Background: CYP26A1 Function and Localization
- CYP26A1 is a cytochrome P450 monooxygenase that belongs to the CYP26 family; it is also known as cytochrome P450RAI or P450RAI1, and plays a critical role in retinoic acid (RA) catabolism.
- This enzyme catalyzes the hydroxylation of all-trans-retinoic acid (atRA) primarily at the C-4 and C-18 positions, initiating the degradation pathway that limits atRA biological activity and controls its homeostasis and signaling.
Related references: PMID:22020119, PMID:9228017, PMID:9716180 - CYP26A1 is also capable of metabolizing other retinoic acid isomers, including 9-cis-RA, 13-cis-RA, and 9,13-di-cis-RA, and can oxidize all-trans-13,14-dihydroretinoate (DRA) to various metabolites.
Related references: PMID:9228017 - It has been shown to contribute to the oxidative metabolism of certain xenobiotics, such as tazarotenic acid.
Related references: PMID:26937021 - CYP26A1 is localized to the membrane of the endoplasmic reticulum and microsomes, consistent with its role as a membrane-bound monooxygenase.
- It is expressed in most fetal and adult tissues, with highest levels in adult liver, heart, pituitary gland, adrenal gland, placenta, and specific brain regions; at the protein level, high expression is observed in lung, pancreas, skin, and uterus.
- The protein contains a heme iron cofactor and is classified under keywords such as alternative splicing, lipid metabolism, monooxygenase, and oxidoreductase.
Experimental Guidance and Technical Tips
- The immunogen maps to amino acids 201–450, which lies within the cytoplasmic catalytic domain of CYP26A1; this design may enhance detection of the native protein across species.
- For Western blotting, a specific band at approximately 56 kDa is expected; positive control lysates from human liver, placenta, or other CYP26A1-expressing tissues are recommended.
- In immunofluorescence and immunohistochemistry, consider antigen retrieval methods optimized for membrane proteins and validate co-localization with ER markers.
- Cross-reactivity with mouse and rat CYP26A1 is indicated, but users should verify species-specific reactivity and antibody titer in their experimental context.
