Biological Background: CYP1B1 Function and Localization
- Cytochrome P450 1B1 (CYP1B1) is a heme-thiolate monooxygenase that metabolizes diverse endogenous substrates, including steroid hormones, fatty acids, and vitamins, using NADPH and molecular oxygen.
Related references: PMID:10681376 PMID:11555828 PMID:12865317 PMID:10426814 - It selectively hydroxylates 17beta-estradiol at the C-4 position, generating 4-hydroxyestradiol, a metabolite implicated in hormonal carcinogenesis.
Related references: PMID:11555828 PMID:12865317 - CYP1B1 converts all-trans retinol to all-trans retinoic acid, playing a key role in extra-hepatic retinoid signaling.
Related references: PMID:10681376 PMID:15258110 - It epoxidizes arachidonic acid to epoxyeicosatrienoic acids (EpETrEs), which act as lipid mediators in the vascular system.
Related references: PMID:20972997 - Additionally, CYP1B1 exhibits NADPH- and O2-independent dehydratase activity toward hydroperoxyeicosatetraenoates (HpETEs).
Related references: PMID:21068195 - Subcellularly, CYP1B1 localizes to the endoplasmic reticulum membrane, microsomal fraction, and mitochondria.
- It is expressed in a broad range of tissues, including heart, brain, lung, kidney, and reproductive organs, and is detected in retinal and umbilical vein endothelial cells.
Related references: PMID:8175734 PMID:19005183 - Mutations in the CYP1B1 gene are associated with primary congenital glaucoma and Peters anomaly, underscoring its importance in ocular development.
Experimental Guidance and Technical Tips
- The immunogen consists of recombinant fusion protein corresponding to the N-terminal region (amino acids 1-260) of human CYP1B1; consider that this antibody may recognize full-length protein and potential isoforms or fragments in lysates.
- For Western blot, a band around 61 kDa is expected; optimization of blocking and incubation conditions is recommended for different sample types.
- In immunofluorescence/immunocytochemistry, appropriate fixation and permeabilization methods should be validated for the target cell type, given CYP1B1's membrane association.
- ELISA applications may require titration of both coating antigen and detection antibody to achieve optimal signal-to-noise ratio.
- Cross-reactivity with mouse and rat CYP1B1 has been observed; sequence homology supports use in these species, but individual validation in the relevant sample system is advised.
