Biological Background: CYP1A1 Function and Localization
- CYP1A1 is a member of the cytochrome P450 superfamily, encoded by the CYP1A1 gene, and is also known as cytochrome P450 form 6, cytochrome P450-C, and cytochrome P450-P1.
- Functions as a monooxygenase that inserts one oxygen atom into substrates, using NADPH and cytochrome P450 reductase for electron transfer, and is involved in the metabolism of fatty acids, steroid hormones, and vitamins. Related references: PMID:10681376, PMID:11555828, PMID:12865317
- Catalyzes the hydroxylation of estrogens, converting estrone and 17beta-estradiol to 2-hydroxy and D-ring hydroxylated metabolites, thereby playing a role in hormone regulation. Related references: PMID:11555828, PMID:12865317, PMID:14559847
- Hydroxylates polyunsaturated fatty acids (PUFAs) with regioselectivity and epoxidates double bonds of PUFAs such as arachidonic acid to generate epoxyeicosatrienoic acids (EETs), which are lipid mediators in the vascular system. Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 261–360 of human CYP1A1, so the antibody is expected to recognize the total protein in denaturing and native applications.
- For Western blotting, an approximate band at 58 kDa should be observed. Validate by including positive controls such as lung or placenta lysates.
- For IHC-P and IF-P, consider optimizing antigen retrieval and validate staining in known expressing tissues (e.g., lung, placenta). Cross-reactivity with mouse and rat indicates a conserved epitope; however, always confirm specificity in the relevant experimental system.
- For ELISA, use appropriate peptide or recombinant protein standards and confirm linearity of detection.
