Biological Background: CYP17A1 Function and Localization
- CYP17A1 (Cytochrome P450 17A1), also known as P450c17, is a member of the cytochrome P450 superfamily encoded by the CYP17A1 gene (aliases: CYP17, S17AH, CPT7).
- Catalyzes both 17-alpha hydroxylase and 17,20 lyase reactions, essential for glucocorticoid and androgen biosynthesis. Converts progesterone and pregnenolone into 17-alpha hydroxy steroids, then cleaves the C17-C20 bond to produce androstenedione and DHEA. Related references: PMID:22266943 PMID:25301938 PMID:27339894
- Demonstrates additional 16-alpha hydroxylase activity, converting 17-alpha hydroxy pregnenolone to 16-alpha-hydroxy DHEA and hydroxylating androgens, contributing to estriol synthesis. Related references: PMID:25301938 PMID:27339894 PMID:36640554
- Subcellularly, CYP17A1 is localized to the endoplasmic reticulum membrane and microsomal membrane.
- Contains a heme iron cofactor and is classified as a monooxygenase/oxidoreductase; it is a phosphoprotein involved in steroid and lipid metabolism.
- Mutations in CYP17A1 are linked to congenital adrenal hyperplasia, a disorder of steroidogenesis.
- The full-length human CYP17A1 protein has a molecular weight of approximately 57kDa.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 209–508 of human CYP17A1) covers part of the catalytic domain, which may confer target specificity.
- In WB, a band near 57kDa is anticipated; verify using positive controls such as adrenal or gonadal tissue lysates.
- Reported cross-reactivity with rat suggests potential utility in rodent models; confirm reactivity in your specific rat sample system.
- For ELISA applications, optimize antibody coating concentrations and consider its use as either capture or detection reagent.
- Because CYP17A1 is an endoplasmic reticulum membrane protein, microsomal or membrane-enriched fractions may improve detection.
- Phosphorylation may cause minor molecular weight shifts; consider phosphatase treatment if precise size confirmation is needed.
