Biological Background: CYP11B2 Function and Localization
- Cytochrome P450 11B2, mitochondrial (aldosterone synthase, ALDOS) is a heme-containing monooxygenase involved in steroid hormone biosynthesis.
- Catalyzes the three-step conversion of 11-deoxycorticosterone to aldosterone, the main mineralocorticoid regulating salt and water homeostasis, blood pressure, and heart failure development. Related references: PMID:11856349 PMID:12530636 PMID:1518866
- Can also produce 18-hydroxycortisol and 18-oxocortisol from cortisol; levels are elevated in primary aldosteronism. Related references: PMID:15356073 PMID:9814482
- Electron transfer from NADPH occurs via adrenodoxin reductase (FDXR) and adrenodoxin (FDX1/FDX2). Related references: PMID:11856349 PMID:1594605 PMID:23322723
- Participates in androgen metabolism by converting testosterone to 11β,17β-dihydroxyandrost-4-ene-3-one. Related references: PMID:23685396
- Can use progesterone as a substrate. Related references: PMID:33189850
- Subcellular location: Mitochondrion inner membrane.
- Tissue specificity: Expressed sporadically in the zona glomerulosa of the adrenal cortex and in aldosterone-producing cell clusters.
- Keywords: Steroidogenesis, oxidoreductase, heme, iron, mitochondrial membrane, disease variant.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (amino acids 1–100). This sequence may be exposed in denaturing conditions, making the antibody suitable for WB. For native detection, IF-P and ELISA may also be attempted, but validation in the relevant sample system is recommended.
- Given the high sequence homology among CYP11B family members, cross-reactivity with other cytochromes (e.g., CYP11B1) might occur. Include isoform-specific positive and negative controls to verify specificity.
- CYP11B2 is a mitochondrial membrane protein. For Western blotting, consider using mitochondrial enrichment or specific lysis buffers to improve detection of this 57 kDa protein.
- The antibody has been tested to cross-react with mouse and rat CYP11B2. When working with these species, validate the antibody performance under your experimental conditions.
