Biological Background: CYLD Function and Localization
-
CYLD is a deubiquitinase that specifically cleaves 'Lys-63'- and linear 'Met-1'-linked polyubiquitin chains, playing a key role in NF-kappa-B activation and TNF-induced necroptosis.

Negatively regulates NF-kappa-B activation by deubiquitinating upstream signaling factors, contributing to the regulation of cell survival, proliferation, and differentiation.

Functions as a negative regulator of Wnt signaling.

Inhibits HDAC6, promoting acetylation of alpha-tubulin and stabilization of microtubules; thereby regulates microtubule dynamics, cell polarization, migration, and angiogenesis.

Required for normal cell cycle progression and cytokinesis.
Related references: PMID:17495026, PMID:19893491 -
Involved in innate immune regulation by restricting linear polyubiquitin formation on RIPK2 and RIPK1, and by modulating NLRP6 inflammasome activity.

Subcellular localization includes cytoplasm, perinuclear region, cytoskeleton, cell membrane, centrosome, spindle, and cilium basal body.
-
Detected in fetal brain, testis, and skeletal muscle; at lower levels in adult brain, leukocytes, liver, heart, kidney, spleen, ovary, and lung. Isoform 2 is found in all tissues except kidney.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 654–953, a region containing part of the USP catalytic domain; this design may confer cross-reactivity across human, mouse, and rat due to sequence conservation.
- For Western blotting, the predicted band is around 107 kDa; ensure adequate gel percentage and transfer conditions for high molecular weight proteins.
- For immunohistochemistry (IHC-P), both nuclear and cytoplasmic staining may be observed, consistent with CYLD localization; optimize antigen retrieval and antibody dilution using appropriate positive control tissues.
- For ELISA, use the recombinant immunogen or target protein for coating and evaluate detection linearity.
- When working with mouse or rat samples, cross-reactivity data suggest applicability; however, validate in the specific tissue or cell context.
