Biological Background: CYLD Function and Localization
- Functions as a deubiquitinase that specifically cleaves 'Lys-63'-linked and linear 'Met-1'-linked polyubiquitin chains. Related references: PMID:18313383, PMID:18636086, PMID:26670046
- Negatively regulates NF-κB signaling by deubiquitinating upstream factors, thereby modulating cell survival, proliferation, and differentiation. Related references: PMID:12917689, PMID:12917691, PMID:32185393, PMID:12917690
- Acts as a negative regulator of the Wnt signaling pathway. Related references: PMID:20227366
- Inhibits HDAC6 to promote α-tubulin acetylation and microtubule stabilization, contributing to cell polarization, migration, and angiogenesis. Related references: PMID:19893491, PMID:18222923, PMID:20194890
- Essential for normal cell cycle progression and cytokinesis. Related references: PMID:17495026, PMID:19893491
- Recruited to the LUBAC complex via SPATA2 to restrict linear ubiquitination on RIPK2 and other targets, thereby regulating innate immune responses and TNF-induced necroptosis. Related references: PMID:26670046, PMID:26997266, PMID:27458237
- Localizes to the cytoplasm, cytoskeleton, centrosome, spindle, and cilium basal body, in line with its roles in microtubule dynamics and cell cycle control.
- Expressed in fetal brain, testis, and skeletal muscle, and at lower levels in adult brain, leukocytes, liver, heart, kidney, spleen, ovary, and lung; isoform 2 is absent in kidney.
Experimental Guidance and Technical Tips
- The immunogen comprising the N-terminal region (aa 1–127) may generate antibodies recognizing the N-terminus of CYLD; validate specificity in your target species and experimental system.
- The antibody is supplied for IF/ICC, IF-P, and ELISA applications; consider optimizing dilution, fixation, and blocking conditions for your specific sample type.
- Cross-reactivity with human and mouse is indicated; when working with other species, verify reactivity experimentally.
- For functional studies involving ubiquitination or signaling, pair with appropriate controls (e.g., deubiquitinase inhibitors or knockdown/knockout validation).
