- CYFIP1 (Cytoplasmic FMR1-interacting protein 1, also known as Sra-1 / p140sra-1) is a 145 kDa adaptor protein encoded by the CYFIP1 gene (GeneID: 23191).
- It is a component of the CYFIP1-EIF4E-FMR1 complex, acting as an adapter between EIF4E and FMR1 to mediate translational repression at the mRNA cap, particularly in the brain.
- CYFIP1 is a core subunit of the WAVE regulatory complex (WRC), which regulates actin filament reorganization via interaction with the Arp2/3 complex; this remodeling activity is controlled by RAC1.
- Through WRC activity, CYFIP1 promotes the formation of membrane ruffles and lamellipodia and plays a role in axon outgrowth and epithelial morphogenesis.
- The protein localizes to the cytoplasm, perinuclear region, cell projections (lamellipodia and ruffles), and is enriched at synapses and synaptosomes.
- It binds F-actin but not RNA, and its actin-remodeling function is critical for cell shape, migration, and differentiation.
