Biological Background: CYFIP1 Function and Localization

  • CYFIP1 (Cytoplasmic FMR1-interacting protein 1, also known as Sra-1 / p140sra-1) is a 145 kDa adaptor protein encoded by the CYFIP1 gene (GeneID: 23191).
  • It is a component of the CYFIP1-EIF4E-FMR1 complex, acting as an adapter between EIF4E and FMR1 to mediate translational repression at the mRNA cap, particularly in the brain.
  • CYFIP1 is a core subunit of the WAVE regulatory complex (WRC), which regulates actin filament reorganization via interaction with the Arp2/3 complex; this remodeling activity is controlled by RAC1.
  • Through WRC activity, CYFIP1 promotes the formation of membrane ruffles and lamellipodia and plays a role in axon outgrowth and epithelial morphogenesis.
  • The protein localizes to the cytoplasm, perinuclear region, cell projections (lamellipodia and ruffles), and is enriched at synapses and synaptosomes.
  • It binds F-actin but not RNA, and its actin-remodeling function is critical for cell shape, migration, and differentiation.
  • CYFIP1 is subject to phosphorylation and alternative splicing, and it is required for BDNF-NTRK2 endocytic trafficking and signaling from early endosomes.
  • It is implicated in neurogenesis and may act as an invasion suppressor in certain cancers.

Experimental Guidance and Technical Tips

  • The immunogen (N-terminal peptide, amino acids 1–100) targets the human CYFIP1 protein; verify reactivity with all desired isoforms in your sample system.
  • In Western blot, a band of approximately 145 kDa is expected; optimize reducing conditions and blocking agents for best signal.
  • For immunofluorescence (IF/ICC), permeabilization and fixation conditions may influence detection of lamellipodial/ruffle localization; consider serum starvation or growth factor stimulation to enhance membrane protrusions.
  • For ELISA applications, the antibody may serve as capture or detection reagent; pair with a validated secondary antibody or matched mAb for sandwich assays.
