Biological Background: Cyclophilin 40 Function and Localization
- Cyclophilin 40, officially designated peptidyl-prolyl cis-trans isomerase D (PPID), is also known as 40 kDa peptidyl-prolyl cis-trans isomerase, CYP-40, and rotamase D. It catalyzes the cis-trans isomerization of proline imidic peptide bonds, a critical step in protein folding.
- As a PPIase, PPID facilitates protein folding by accelerating the isomerization of proline bonds in oligopeptides. Related references: PMID:11350175 PMID:20676357
- PPID acts as a co-chaperone within HSP90 complexes, particularly in unligated steroid receptor heterocomplexes. It shows preference for estrogen receptor complexes and may influence receptor trafficking via cytoplasmic dynein-dependent movement to the nucleus.
- Beyond its chaperone role, PPID inhibits the DNA-binding activity of MYB, regulates AHR signaling by promoting AHR:ARNT dimer formation independently of HSP90, and modulates UV radiation-induced apoptosis. It also promotes cell viability in ALK+ anaplastic large-cell lymphoma (ALCL) cell lines.
- PPID may be involved in the replication and release of hepatitis C virus (HCV).
- Subcellularly, PPID localizes to the cytoplasm, nucleus, nucleolus, and nucleoplasm.
- PPID is widely expressed across tissues.
- Post-translational modifications include acetylation and phosphorylation, and the protein contains TPR repeats characteristic of immunophilins.
Experimental Guidance and Technical Tips
- For Western blotting, use appropriate loading controls and validate the expected band size of approximately 41 kDa. Since the antibody is polyclonal and cross-reacts with rat, consider validating in your specific sample system.
- In ELISA, the antibody can be used as a detection or capture reagent depending on your assay design. Optimize dilution factors empirically.
- Given the polyclonal nature, this antibody may detect multiple epitopes on Cyclophilin 40, potentially enhancing signal in denaturing and native conditions.
