Biological Background: Cyclin T1 Function and Localization
- Cyclin T1 (CCNT1) is the regulatory subunit of the positive transcription elongation factor b (P-TEFb) complex, which it forms with cyclin-dependent kinase 9 (CDK9). Related references: PMID:16109376 PMID:16109377 PMID:30134174
- P-TEFb phosphorylates the C-terminal domain (CTD) of RNA polymerase II, enabling the transition from abortive to productive transcription elongation. Related references: PMID:16109376 PMID:16109377 PMID:30134174
- Cyclin T1 promotes liquid-liquid phase separation (LLPS) that enhances P-TEFb binding to the RNA polymerase II CTD. Related references: PMID:29849146 PMID:35393539
- During HIV/SIV infection, Cyclin T1 binds the viral Tat protein and the transactivation response element (TAR), acting as an essential cofactor for viral gene transcription.
- Encoded by the CCNT1 gene, Cyclin T1 is a member of the cyclin family and contains a characteristic cyclin box domain.
- The protein localizes to the nucleus and undergoes multiple post‑translational modifications including acetylation, phosphorylation, and ubiquitination.
- Cyclin T1 is ubiquitously expressed across human tissues.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region (aa 220–320) of human Cyclin T1; therefore, the antibody may recognize full-length Cyclin T1 as well as isoforms containing this sequence.
- For Western blot (WB), expect a band at approximately 81 kDa in human cell or tissue lysates. Consider using nuclear extracts for enriched signal and include appropriate positive/negative controls.
- For ELISA, this polyclonal antibody may be paired with a detection antibody; titrate both capture and detection antibodies to optimize assay sensitivity and specificity.
- Validate the antibody in the relevant human sample system under the specific conditions of your experiment, as reactivity may vary with sample preparation and epitope accessibility.
