Biological Background: CYC1 Function and Localization
- CYC1 encodes cytochrome c1, a heme protein that is part of the mitochondrial respiratory chain complex III (ubiquinol-cytochrome c oxidoreductase). It is also known as Complex III subunit 4, UQCR4, and MC3DN6.
- Cytochrome c1 is a catalytic core subunit containing a c-type heme group that transfers electrons from the Rieske iron-sulfur protein to cytochrome c.
- The cytochrome b-c1 complex catalyzes the transfer of electrons from ubiquinol to cytochrome c, coupling this redox reaction to proton translocation across the mitochondrial inner membrane via the Q cycle.
- Located in the mitochondrial inner membrane, CYC1 is a single-pass transmembrane protein with a transit peptide for mitochondrial import.
- Cytochrome c1 is subject to phosphorylation, which may regulate its activity or assembly.
- Mutations in CYC1 are associated with primary mitochondrial disease, specifically mitochondrial complex III deficiency, nuclear type 6 (MC3DN6).
- The mature protein has a molecular weight of approximately 35 kDa (35422 Da from UniProt, consistent with observed 35 kDa on SDS-PAGE).
Experimental Guidance and Technical Tips
- The antibody is raised against a recombinant protein spanning amino acids 85-291 of human CYC1 (NP_001907.2). This immunogen covers the mature peptide after removal of the mitochondrial targeting sequence, and includes the heme-binding and transmembrane domains.
- For Western blotting, a predominant band at approximately 35 kDa is expected in human, mouse, and rat mitochondrial extracts. Validate loading controls and consider using protease inhibitors to prevent degradation.
- In immunohistochemistry (IHC-P), antigen retrieval may be required; test both heat-induced and enzymatic methods. For ELISA, direct coating of the recombinant immunogen or purified complex III may be considered as a positive control.
