Biological Background: CYBA (p22phox) Function and Localization

  • CYBA encodes the p22phox protein, the light chain of the cytochrome b-245 complex (also known as cytochrome b(558)), which is an essential subunit of the phagocyte NADPH oxidase complex. Related references: PMID:15824103, PMID:17140397, PMID:38355798

  • The NADPH oxidase complex transfers electrons from cytosolic NADPH to molecular oxygen, generating the superoxide anion (O2(-)), a reactive oxygen species crucial for host defense and cell signaling. Related references: PMID:38355798

  • p22phox acts as a scaffold for the assembly of cytosolic regulatory subunits, and its C-terminal region is phosphorylated, allowing binding of NCF1/p47-phox to initiate complex activation. Related references: PMID:19948736

  • CYBA also associates with NOX3 to form a constitutively active NADPH oxidase that generates superoxide in non-phagocytic cells. Related references: PMID:15824103, PMID:17140397

  • The protein localizes to the cell membrane and phagosomal membrane upon activation.

  • CYBA contains transmembrane helices and binds heme and iron cofactors essential for electron transfer.

  • Mutations in CYBA are associated with chronic granulomatous disease (CGD), a primary immunodeficiency characterized by defective superoxide production.

  • The protein is subject to post-translational modifications including phosphorylation and ubiquitination.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 126–195 of human CYBA, a region that is conserved across human, mouse, and rat, consistent with the observed cross-reactivity. For detection of endogenous CYBA, consider validating the antibody in relevant cell lines (e.g., neutrophils, macrophages) or tissues.
  • For western blotting, a predicted molecular weight of 21 kDa is expected. However, post-translational modifications may cause slight shifts. Include appropriate positive and negative controls.
  • In immunofluorescence applications, CYBA localizes to the plasma membrane, so membrane staining patterns should be observed. Permeabilization conditions must be optimized for the target cell type.
  • ELISA protocols should be optimized with a suitable coating buffer and detection system, ensuring proper blocking to reduce nonspecific binding.
