Biological Background: CXCR3 Function and Localization
- CXCR3, also known as GPR9 or CD183, is a G protein-coupled receptor encoded by the CXCR3 gene. Alternative splicing produces at least two isoforms with distinct tissue expression profiles.
- It functions as a receptor for the C-X-C chemokines CXCL9, CXCL10, and CXCL11, mediating proliferation, survival, and angiogenic activity in human mesangial cells through heterotrimeric G-protein signaling, and promoting chemotaxis via Ras/ERK pathway activation. Related references: PMID:12782716
- CXCR3 also binds CXCL4, mediating anti-proliferative and anti-angiogenic effects in human microvascular endothelial cells through a cAMP-dependent pathway and p38MAPK activation. Overexpression in renal cancer cells downregulates HMOX1 and promotes apoptosis. Related references: PMID:12782716
- The receptor mediates the specific activity of CXCL11.
- Subcellularly, CXCR3 is localized to the cell membrane.
