Biological Background: CXCL9 Function and Localization
- CXCL9, also known as monokine induced by interferon-gamma (MIG), is a member of the CXC chemokine family and contains conserved cysteine residues that form disulfide bonds essential for its structure.
- It functions as a cytokine that affects the growth, movement, and activation of immune cells, acting as a potent chemoattractant for activated T lymphocytes.
- CXCL9 mediates its effects by binding to the chemokine receptor CXCR3, a G protein-coupled receptor expressed on activated T cells and other immune cells.
- The protein is secreted and plays a key role in inflammatory responses, with expression strongly induced by interferon-gamma.
- The mature protein has a calculated molecular mass of approximately 14 kDa, consistent with its small cytokine nature.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 50 to the C-terminus of human CXCL9; epitope mapping may be considered if detailed binding characteristics are required.
- For Western blot, a band near 14 kDa is expected; validate antibody performance in appropriate positive control samples such as stimulated immune cells or tissues expressing CXCL9.
- In ELISA applications, this antibody may be used for capture or detection; however, optimal pairing with a suitable monoclonal antibody or recombinant antigen is recommended for sandwich formats.
- Cross-reactivity with human CXCL9 is confirmed; reactivity with other species or related chemokines should be independently tested.
