Biological Background: CXCL4/PF4 Function and Localization
- Platelet factor 4 (PF4), also known as C-X-C motif chemokine 4, Iroplact, or Oncostatin-A, is a secreted chemokine primarily released during platelet aggregation.
- It functions in various biological processes including hematopoiesis, cell proliferation, differentiation, and activation, acting through receptors CCR1, CXCR3A, and CXCR3B. Related references: PMID:29930254 PMID:9531587
- Upon binding to CXCR3A, PF4 induces migration of activated T-lymphocytes via downstream Ras/ERK signaling, contributing to inflammatory responses. Related references: PMID:18174362 PMID:24469069
- PF4 neutralizes the anticoagulant effect of heparin by competitively binding to heparin, and it enhances protein C activation through interaction with thrombomodulin. Related references: PMID:9395524
- It inhibits hematopoiesis and maintains hematopoietic stem cell quiescence, and also acts as a chemotactic factor for neutrophils and monocytes via CCR1. Related references: PMID:9531587 PMID:29930254
- In endothelial cells, PF4 inhibits proliferation and promotes endothelial-to-mesenchymal transition, leading to myofibroblast differentiation and collagen synthesis. Related references: PMID:34986347
- In cooperation with toll-like receptor 8 (TLR8), PF4 induces chromatin remodeling and activates inflammatory gene expression via the TBK1-IRF5 axis. Related references: PMID:35701499
- Post-translational modifications include phosphorylation and disulfide bond formation; it is a well-characterized reference proteome member with a known 3D structure.
Experimental Guidance and Technical Tips
- The immunogen sequence (amino acids 32–101) corresponds to the mature PF4 protein, suggesting the antibody recognizes the secreted form. For WB, a band around 11 kDa is expected under reducing conditions; multi-band patterns may arise from dimerization or complex formation with heparin.
- PF4 is a secreted cytokine, so consider testing cell culture supernatants, serum, or plasma samples. For tissue staining (IHC-P, IF-P), optimize fixation and antigen retrieval protocols, as PF4 may be bound to extracellular matrix or cell-surface glycosaminoglycans.
- The antibody cross-reacts with mouse and rat PF4, but validate specificity in your model system due to potential sequence variations among species.
- As a polyclonal antibody, performance may vary between batches; consistent QC evaluation in your target application is advised.
