Biological Background: CXCL2 Function and Localization
- CXCL2, also designated Gro-beta or MIP2-alpha, is a secreted C-X-C motif chemokine that plays a central role in the inflammatory response.
- It is produced by activated monocytes and neutrophils and functions as a chemoattractant, directing immune cells to sites of inflammation.
- In vitro, CXCL2 acts as a hematoregulatory molecule, suppressing hematopoietic progenitor cell proliferation.
- The mature protein is proteolytically processed to yield the bioactive fragment GRO-beta(5-73), which exhibits markedly enhanced hematopoietic activity.
- Like other C-X-C chemokines, CXCL2 contains conserved disulfide bonds that stabilize its tertiary structure.
- Subcellularly, CXCL2 is secreted into the extracellular milieu to mediate intercellular signaling.
- The predicted molecular weight of the unprocessed precursor is approximately 11.4 kDa (UniProt P19875).
Experimental Guidance and Technical Tips
- This rabbit polyclonal antibody was raised against the full-length human CXCL2 (residues 1–107) and is expected to recognize both the precursor and mature forms.
- For Western blotting, a specific band around 11 kDa should be observed; consider using lysates from activated monocyte or neutrophil cell lines as positive controls.
- The antibody cross-reacts with rat CXCL2, enabling its use in rat models of inflammation.
- In ELISA applications, titration of both antibody and antigen is recommended to achieve optimal signal-to-noise ratios.
- As with any polyclonal reagent, lot-to-lot consistency should be confirmed for quantitative assays.
