Biological Background: CXCL12 Function and Localization
- CXCL12, also known as stromal cell-derived factor 1 (SDF-1), is a secreted CXC chemokine that exists as multiple isoforms (alpha, beta, gamma, delta, epsilon, theta) due to alternative splicing.
- CXCL12 functions as a chemoattractant for T-lymphocytes and monocytes, but not neutrophils, by binding and activating the CXCR4 receptor, leading to rapid intracellular calcium mobilization and directed cell migration. Related references: PMID:18802065, PMID:39093700, PMID:8752281
- It also binds to the atypical chemokine receptor ACKR3 (CXCR7), which serves as a scavenger receptor and activates the beta-arrestin signaling pathway. Related references: PMID:16107333, PMID:19255243
- CXCL12 can activate integrins ITGAV:ITGB3, ITGA4:ITGB1, and ITGA5:ITGB1 via an allosteric binding site, independent of CXCR4. Related references: PMID:29301984

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 40–93 of human CXCL12 (NP_000600.1), a region within the mature chemokine domain; therefore, the antibody is expected to recognize the secreted protein. For Western blot, prepare samples under reducing conditions and use a standard SDS-PAGE protocol. Consider a starting dilution range of 1:500–1:2000, followed by optimization.
- For IHC-P, perform heat-induced antigen retrieval and validate staining with appropriate tissue controls. CXCL12 is secreted, so extracellular staining patterns may be observed.
- In IF/ICC, consider using a secretion inhibitor (e.g., brefeldin A) to enhance intracellular signal; optimize fixation/permeabilization conditions. The antibody may label the Golgi/secretory pathway.
- The antibody cross-reacts with mouse and rat CXCL12, making it suitable for studies in these species. However, always confirm specificity in the relevant model system.
- Due to alternative splicing, multiple bands may be detected in WB; compare with known molecular weights of isoforms (alpha ~11 kDa). Use a positive control (e.g., recombinant CXCL12 or cell lines expressing high levels) to verify reactivity.
