Biological Background: CXCL12 Function and Localization
- CXCL12 (Stromal cell-derived factor 1) is a chemokine of the C-X-C family, with multiple isoforms (Alpha, Beta, Gamma, Delta, Epsilon, Theta) generated by alternative splicing.
- It functions as a chemoattractant for T-lymphocytes and monocytes, but not neutrophils, signaling through CXCR4 to induce calcium flux and chemotaxis. Related references: PMID:8752281, PMID:18802065, PMID:39093700
- CXCL12 also binds the atypical receptor ACKR3, activating beta-arrestin pathways, acting as a scavenger receptor, and inducing down-regulation and internalization of ACKR3. Related references: PMID:16107333, PMID:19255243
- Through an allosteric site on integrins, CXCL12 can activate ITGAV:ITGB3, ITGA4:ITGB1, and ITGA5:ITGB1 independently of CXCR4. Related references: PMID:29301984
