Biological Background: CXCL10 Function and Localization
- CXCL10, also known as IP-10 or small-inducible cytokine B10, is a pro-inflammatory chemokine that regulates chemotaxis, differentiation, activation of immune cells, cell growth, apoptosis, and angiostatic effects. Related references: PMID:11157474 PMID:22652417 PMID:7540647
- It plays a critical role during viral infections by stimulating immune cell activation and migration to infected sites (By similarity).
- CXCL10 exerts its effects by binding to the CXCR3 receptor, activating G protein-mediated signaling, which increases intracellular calcium and induces actin reorganization. Related references: PMID:12750173 PMID:19151743
- This signaling cascade recruits activated Th1 lymphocytes to inflammation sites, directing the adaptive immune response. Related references: PMID:12663757 PMID:12750173
- In the central nervous system, activation of the CXCL10/CXCR3 axis in neurons following brain injury helps to activate and direct microglia to the lesion site, facilitating neuronal reorganization (By similarity).
- CXCL10 is secreted and mainly produced by monocytes, endothelial cells, fibroblasts, thymic epithelial cells, and microglia in response to interferon-gamma and viral stimuli. Related references: PMID:11157474 PMID:12663757
- Key structural and functional features include chemotaxis, inflammatory response, and disulfide bond formation, characteristic of the CXC chemokine family.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 22-98 of mature CXCL10, which excludes the signal peptide and covers the full chemokine domain; this region is expected to produce antibodies recognizing the native secreted form.
- For Western blot, the predicted molecular weight is approximately 11 kDa; use reducing SDS-PAGE conditions and validate the band in samples known to express CXCL10 after cytokine stimulation.
- In immunofluorescence or immunocytochemistry, because CXCL10 is secreted, consider treating cells with a Golgi-blocking agent (e.g., brefeldin A) before fixation to enhance intracellular staining, and validate in cell types such as IFN-γ-treated fibroblasts or monocytes.
- For ELISA applications, the antibody can be evaluated as a capture or detection reagent in a sandwich ELISA format, but optimal pairing with a matched antibody should be experimentally determined.
- Since the antibody shows cross-reactivity with mouse, it may be suitable for studies in mouse models; however, confirm specificity in the relevant sample system.
