Biological Background: C-X-C motif chemokine 10 (CXCL10) Function and Localization
- C-X-C motif chemokine 10 (CXCL10), also known as 10 kDa interferon gamma-induced protein (Gamma-IP10, IP-10) or Small-inducible cytokine B10, is a secreted cytokine encoded by the CXCL10 gene (Gene ID: 3627).
- Functions as a pro-inflammatory cytokine, regulating chemotaxis, differentiation, and activation of peripheral immune cells, as well as cell growth, apoptosis, and angiostatic effects [PMID:11157474, PMID:22652417, PMID:7540647]. Related references: PMID:11157474 PMID:22652417 PMID:7540647
- Binds to the CXCR3 receptor, activating G protein-mediated signaling that increases intracellular calcium and leads to actin reorganization, ultimately promoting chemotaxis of activated Th1 lymphocytes to inflammation sites [PMID:12750173, PMID:19151743, PMID:12663757]. Related references: PMID:12750173 PMID:19151743 PMID:12663757
- In the central nervous system, the CXCL10/CXCR3 axis mediates microglial activation and recruitment following brain injury, which is essential for neuronal reorganization (By similarity).
- Mainly secreted by monocytes, endothelial cells, and fibroblasts; also expressed in thymic epithelial cells and microglial cells upon viral stimulation [PMID:11157474, PMID:12663757]. Related references: PMID:11157474 PMID:12663757
- Contains a signal peptide and a disulfide bond, and is subject to citrullination; classified under inflammatory response, chemotaxis, and secreted proteins (UniProt keyword).
Experimental Guidance and Technical Tips
- For Western blotting, the expected molecular weight is approximately 11 kDa; the immunogen corresponds to the mature peptide (aa 20-98) without the signal peptide, so detection of secreted forms may require non-reducing conditions or careful sample preparation.
- As a secreted cytokine, CXCL10 is best analyzed in conditioned media, serum, or tissue extracts from activated immune cells; consider inducing expression with interferon-gamma or viral stimuli in appropriate cell lines.
- For ELISA applications, pairing this polyclonal antibody as either capture or detector with a well-characterized monoclonal can improve specificity; validate in the relevant sample matrix.
