Biological Background: Fractalkine (CX3CL1) Function and Localization

  • CX3CL1 (Fractalkine) is a C-X3-C motif chemokine that acts as a ligand for CX3CR1 and integrins ITGAV:ITGB3 and ITGA4:ITGB1, mediating immune response, inflammation, cell adhesion, and chemotaxis.
    Related references: PMID:10187784 PMID:18971423
  • Through CX3CR1 signaling, CX3CL1 regulates leukocyte adhesion and migration at the endothelium, and can activate integrins in both a CX3CR1-dependent and -independent manner.
    Related references: PMID:10382755 PMID:9177350
  • The membrane-bound form promotes adhesion of leukocytes to endothelial cells, while the proteolytically released soluble form is chemotactic for T-cells and monocytes, but not neutrophils.
  • Fractalkine is predominantly expressed in the brain, with lower levels in kidney, heart, lung, skeletal muscle, and testis; it is also found in dendritic cells and stimulated B‑cells.
    Related references: PMID:10508268 PMID:9177350 PMID:9479488 PMID:9845323 PMID:12569158
  • The protein is a type I transmembrane protein containing a signal peptide, a mucin‑like stalk, a transmembrane helix, and a short cytoplasmic tail; it can be cleaved to release the soluble chemokine domain.
  • The chemokine domain is stabilized by conserved disulfide bonds, consistent with its classification in the C‑X3‑C subfamily.

Experimental Guidance and Technical Tips

  • The immunogen (aa 25–337) spans the extracellular chemokine domain and part of the mucin stalk; the antibody may detect full‑length (~42 kDa) and cleaved/shed forms in WB.
  • For IHC‑P and IF‑P, expect membrane‑localized staining; optimize fixation and permeabilization conditions for mouse tissues or cell lines.
  • In ELISA, the antibody can be used to capture or detect soluble fractalkine in cell culture supernatants or biological fluids; validate in the relevant sample matrix.
  • As a polyclonal antibody, lot‑to‑lot variability may exist; consider including appropriate controls and routine validation in your assay system.
