- CUX1 (Homeobox protein cut-like 1), also known as CDP (CCAAT displacement protein) and CDP/Cux p200, is a transcription factor with multiple isoforms generated through alternative splicing and proteolytic processing.
- It plays a central role in neuronal differentiation in the brain, regulating dendrite development, branching, and dendritic spine formation in cortical layers II–III, and also controls synaptogenesis.
- Acts as a transcriptional repressor by binding to CCAAT motifs, preventing positively acting CCAAT factors from binding; it is a component of the nf-munr repressor and binds to matrix attachment regions (MARs) of the immunoglobulin heavy chain enhancer.
- Represses T-cell receptor beta enhancer function by binding to MARbeta and may require TCF4 as a coactivator for other targets.
- CUX1 is involved in cell cycle progression at the G1/S transition; as cells enter S phase, proteolytic processing generates a stable, DNA-binding p110 fragment that activates genes like POLA1.
- Subcellular localization is primarily nuclear, consistent with its role as a transcription factor.
- The protein is subject to phosphorylation and ubiquitination (isopeptide bond formation), and mutations are linked to intellectual disability.
