Biological Background: Cullin-4A Function and Localization

  • CUL4A serves as a core scaffold protein within cullin-RING E3 ubiquitin ligase complexes (CRLs), enabling ubiquitin transfer to substrate proteins by positioning both the substrate and the ubiquitin-conjugating enzyme. The ligase activity is dependent on cullin neddylation and is inhibited by association of deneddylated CUL4A with CAND1. Related references: PMID:14578910, PMID:14739464, PMID:15448697
  • The DCX(DDB2) complex employs DDB2 as a substrate receptor to ubiquitinate XPC, histones H3-H4, and other targets, playing critical roles in nucleotide excision repair and replication-coupled nucleosome assembly. Related references: PMID:15811626, PMID:16678110, PMID:17041588, PMID:24209620
  • Through the DCX(DTL) complex, CUL4A mediates PCNA-dependent polyubiquitination of CDT1 and MDM2-dependent ubiquitination of p53/TP53 in response to DNA damage and during DNA replication, and also directs autoubiquitination of DTL. Additionally, the DDB1-CUL4A-DTL complex regulates circadian rhythm by ubiquitinating CRY1 for degradation. Related references: PMID:14578910, PMID:15448697, PMID:15548678, PMID:23478445, PMID:26431207
  • The DCX(ERCC8) complex (CSA complex) participates in transcription-coupled repair (TCR), removing DNA lesions from actively transcribed genes. Related references: PMID:12732143, PMID:32355176, PMID:38316879
  • The DCX(AMBRA1) complex functions as a master regulator of the G1/S cell cycle transition by targeting phosphorylated cyclin D (CCND1, CCND2, CCND3) for ubiquitination and degradation. It also regulates CRL5 complexes through ubiquitination and degradation of Elongin-C (ELOC). Related references: PMID:33854232, PMID:33854239, PMID:30166453
  • CUL4A is implicated in ubiquitination of numerous other substrates, including JUN (via DET1-COP1), CDKN1B/p27 (with DDB1-SKP2), and HOXA9, highlighting its broad regulatory roles. Related references: PMID:14739464, PMID:16537899, PMID:14609952
  • Together with CUL4B, CUL4A contributes to ribosome biogenesis. Related references: PMID:26711351

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 1–300 of human CUL4A, indicating that the antibody targets the N-terminal region; this may influence epitope accessibility in certain assays.
  • In Western blotting (WB), expect a band at approximately 88 kDa; include protease inhibitors during lysis and consider using fresh samples to avoid degradation.
  • For immunoprecipitation (IP), validate the antibody’s ability to capture native CUL4A complexes under non-denaturing conditions.
  • For immunofluorescence (IF/ICC), optimize fixation/permeabilization protocols, as CUL4A is predominantly nuclear; confirm staining specificity with appropriate controls.
  • This antibody cross-reacts with human, mouse, and rat CUL4A; when using rodent samples, ensure that the detection system is compatible and that the target band is distinguishable from endogenous immunoglobulins in IP.
  • As a polyclonal antibody, lot-to-lot variability may occur; consider titrating the antibody for each new application and including positive and negative controls.
