Biological Background: Cullin-1 Function and Localization
- Core component of multiple cullin-RING-based SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complexes, which mediate the ubiquitination of proteins involved in cell cycle progression, signal transduction and transcription. - In the SCF complex, serves as a rigid scaffold that organizes the SKP1-F-box protein and RBX1 subunits. May contribute to catalysis through positioning of the substrate and the ubiquitin-conjugating enzyme. Related references: PMID:38326650
- The E3 ubiquitin-protein ligase activity of the complex is dependent on the neddylation of the cullin subunit and exchange of the substrate recognition component is mediated by TIP120A/CAND1. Related references: PMID:12609982, PMID:38326650
- The functional specificity of the SCF complex depends on the F-box protein as substrate recognition component. For example, SCF(SKP2) directs ubiquitination of p27 to regulate G1/S transition, while SCF(BTRC) targets β-catenin in Wnt signaling.

Experimental Guidance and Technical Tips
- The immunogen spans amino acids 435–593 (C-terminal region). In Western blotting, expect a band around 90 kDa; validate specificity using positive controls such as lung fibroblast lysates.
- For IHC-P, consider standard antigen retrieval protocols and include a known positive tissue (e.g., human lung section) to confirm staining.
- For IF/ICC, optimize fixation and permeabilization conditions; Cullin-1 localizes to both nucleus and cytoplasm.
- In ELISA, this antibody can be used for capture or detection; pair with a matched antibody set for optimal sensitivity.
