Biological Background: CUL5 Function and Localization

  • CUL5 encodes the scaffold protein cullin-5 (VACM-1) that forms the core of multiple cullin-5-RING E3 ubiquitin ligase complexes (CRL5), positioning substrates and ubiquitin-conjugating enzymes for ubiquitination and proteasomal degradation.   • The substrate specificity of CRL5 is conferred by SOCS box proteins; for instance, ECS(SOCS1) ubiquitinates JAK2, while ECS(SOCS2) targets phosphorylated EPOR and GHR. Related references: PMID:11384984, PMID:21980433, PMID:25505247

  • CUL5 regulates neuron positioning during cortex development by mediating DAB1 degradation via SOCS7-containing ECS complexes (By similarity).
