Biological Background: CUL4B Function and Localization
- CUL4B functions as a scaffold protein within cullin-RING E3 ubiquitin ligase complexes, orchestrating substrate positioning and ubiquitin transfer to target proteins for proteasomal degradation. Related references: PMID:14578910, PMID:16678110, PMID:18593899
- In response to DNA damage, CUL4B-containing complexes mediate polyubiquitination of CDT1 and histones H2A, H3, and H4, and are targeted to UV-damaged chromatin via DDB2, implicating CUL4B in DNA repair and replication. Related references: PMID:14578910, PMID:16678110, PMID:18593899
- The DCX(AMBRA1) subcomplex, containing CUL4B, acts as a master regulator of G1/S transition by ubiquitinating phosphorylated cyclins D1, D2, and D3, and also modulates Cul5-RING ligases via degradation of Elongin-C. Related references: PMID:33854232, PMID:33854239, PMID:30166453
- CUL4B is required for ubiquitination of cyclin E (CCNE1/CCNE2) and ensures normal G1 progression, highlighting its critical role in cell cycle control. Related references: PMID:16322693, PMID:19801544
- It regulates the mTORC1 signaling pathway in a proteasome-dependent manner through interaction with MLST8, influencing cell growth, size, and metabolism. Related references: PMID:18235224
- Together with CUL4A, CUL4B contributes to ribosome biogenesis, underscoring its broader role in cellular anabolic processes. Related references: PMID:26711351
- CUL4B localizes to both the cytoplasm and nucleus and undergoes post-translational modifications including acetylation and phosphorylation.
- Mutations in CUL4B are associated with X-linked intellectual disability, short stature, and other developmental abnormalities.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal 1-152 amino acids of human CUL4B; this region may be conserved across species, consistent with the observed human/mouse cross-reactivity.
- For Western blot, use standard reducing conditions and a loading control (e.g., GAPDH) to validate specificity; the expected band is ~104 kDa.
- Optimize primary antibody dilution and incubation conditions (e.g., 1:500–1:2000 for WB) using positive cell lysates such as HeLa or HEK293, and always include a negative control (e.g., knockdown lysates if available).
- In ELISA, consider coating the recombinant immunogen or purified protein, and titrate both capture and detection antibody concentrations to achieve optimal signal-to-noise ratios.
