Biological Background: CTP synthase / CTPS Function and Localization
- CTP synthase 1 (CTPS1, also designated CTP synthetase 1, UTP--ammonia ligase 1, and GATD5) is a 67 kDa enzyme encoded by the CTPS1 gene (GeneID: 1503) and serves as the key regulatory enzyme in de novo CTP biosynthesis.
- CTPS1 catalyzes the ATP-dependent amination of UTP to CTP, a critical precursor for DNA, RNA, and phospholipid synthesis. This activity is essential for cellular proliferation and viability. Related references: PMID:16179339 PMID:17189248 PMID:17463002
- CTPS1 activity is particularly crucial for the proliferation of activated lymphocytes, supporting the increased demand for nucleotides and lipid membranes during immune responses. Defects in CTPS1 are associated with immunodeficiency (IMD24). Related references: PMID:24870241 PMID:8530356
- Beyond its ligase function, CTPS1 acts as a protein-asparagine deamidase, removing the side chain amide group from specific asparagine residues to generate aspartate, thereby regulating protein function. Related references: PMID:40240600
- CTPS1 negatively regulates innate immunity by deamidating 'Asn-85' of IRF3, impairing its DNA-binding ability and downstream interferon signaling.
- In response to DNA damage, CTPS1 deamidates histone H1 at Asn-76 and Asn-77, promoting H1K75 acetylation and chromatin relaxation to facilitate the recruitment of DNA repair proteins. Related references: PMID:40240600
- CTPS1 is primarily localized in the cytosol and cytoplasm, but also translocates to the nucleus and associates with chromosomes, consistent with its dual roles in nucleotide metabolism and chromatin regulation.
- CTPS1 is ubiquitously expressed at low levels in most tissues, with a notable upregulation in activated T-lymphocytes, reflecting its importance in immune cell proliferation.
Experimental Guidance and Technical Tips
- The immunogen was designed against a conserved region (aa 200–300) of human CTPS1, which shares high homology with mouse and rat orthologs, supporting the observed cross-reactivity.
- For Western blot, anticipate a band near 67 kDa; optimize gel percentage and transfer conditions for this molecular weight range.
- The antibody can be applied in immunoprecipitation (IP) and ELISA; consider titrating the antibody for each assay and including appropriate controls.
- Validate the antibody’s performance in the relevant sample system—such as activated T-cell lysates or CTPS1-overexpressing cell lines—to confirm specificity and sensitivity.
