Biological Background: Catenin delta-2 Function and Localization
- Catenin delta-2 (CTNND2), also known as Delta-catenin, GT24, Neural plakophilin-related ARM-repeat protein (NPRAP), and Neurojungin, is encoded by the CTNND2 gene (synonym NPRAP).
- Plays a critical role in neuronal development, particularly in the formation and/or maintenance of dendritic spines and synapses. Related references: PMID:25807484
- Involved in the regulation of Wnt signaling by modulating beta-catenin turnover; facilitates beta-catenin interaction with GSK3B, leading to its phosphorylation, ubiquitination, and degradation (By similarity). Related references: PMID:25807484
- Functions as a transcriptional activator when bound to ZBTB33 (By similarity).
- May contribute to neuronal cell adhesion and tissue morphogenesis through regulation of adhesion molecules.
- Localizes to the nucleus, adherens junctions, dendrites, and perikaryon.
- Highly expressed in brain, with the highest levels observed in fetal brain. Related references: PMID:25807484
- Post-translational modifications include glycosylation, phosphorylation, and methylation; associated with autism spectrum disorder (ASD) and other disease variants.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 966-1225 of human CTNND2, which may detect full-length protein and certain isoforms; verify specificity in your experimental system.
- For Western blot, expect a band around 133 kDa; consider using a low-percentage SDS-PAGE gel and extended transfer times for efficient high molecular weight protein detection.
- The antibody has been validated for ELISA; optimize coating and detection conditions for your specific sample matrix.
- Cross-reactivity with human and rat has been reported; for other species, validation is recommended before use.
