Biological Background: CTNNBL1 Function and Localization

  • CTNNBL1, also known as Beta-catenin-like protein 1, NAP, or NYD-SP19, is a nuclear and cytoplasmic protein encoded by the CTNNBL1 gene (C20orf33) in humans.
  • It is a component of the PRP19-CDC5L complex, an integral part of the spliceosome essential for pre-mRNA splicing activation.
  • CTNNBL1 participates in AID/AICDA-mediated somatic hypermutation (SHM) and class-switch recombination (CSR), processes that generate high-affinity, isotype-switched antibodies. Related references: PMID:32484799
  • The protein localizes to both the nucleus and cytoplasm, consistent with its roles in splicing and immune diversification.
  • It is widely expressed, with highest levels in skeletal muscle, placenta, heart, spleen, testis, and thyroid.
  • Annotated keywords include alternative splicing, adaptive immunity, phosphoprotein, acetylation, and disease variant, underscoring its dynamic regulation and clinical relevance.
  • The full-length protein has a molecular weight of approximately 65 kDa (UniProt: 65173 Da).

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 264–563, a C-terminal region of CTNNBL1; the resulting polyclonal antibody may recognize multiple epitopes and isoforms.
  • For Western blot, expect a band around 65 kDa in human, mouse, and rat lysates. Validate in positive tissues such as testis or spleen that express high levels.
  • In immunofluorescence/ICC, CTNNBL1 localizes to the nucleus and cytoplasm. Use a nuclear counterstain to confirm subcellular distribution.
  • For ELISA, optimize coating antigen and antibody dilutions using the recombinant immunogen or a relevant peptide.
  • Cross-reactivity with mouse and rat has been observed; however, verify sequence homology in the immunogen region for other species if needed.
