Biological Background: CTIP2/BCL11B Function and Localization

  • CTIP2/BCL11B (B-cell lymphoma/leukemia 11B) is a zinc-finger transcription factor that acts as a key regulator of T-lymphocyte differentiation and survival during thymocyte development. It controls the responsiveness of hematopoietic stem cells to chemotactic signals by modulating CCR7 and CCR9 expression, directing progenitor cell migration from bone marrow to thymus. Related references: PMID:27959755
  • It enhances IL2 expression in activated CD4+ T-lymphocytes, functioning as a transcriptional regulator of the IL2 promoter. Related references: PMID:16809611
  • CTIP2/BCL11B is a tumor suppressor that represses transcription through direct, TFCOUP2-independent binding to a GC-rich response element. It may also participate in the P53-signaling pathway.
  • Multiple isoforms are produced by alternative splicing, including ATL1-alpha, ATL1-beta, ATL1-delta, ATL1-gamma, and hRIT1-alpha.
  • The protein localizes to the nucleus, consistent with its role as a transcription factor.
  • It is highly expressed in the brain and in malignant T-cell lines derived from patients with adult T-cell leukemia/lymphoma.
  • Key post-translational modifications include acetylation, methylation, phosphorylation, and ubiquitination. It also features isopeptide bonds and metal-binding zinc-finger domains.
  • Associated with disease variants linked to intellectual disability and severe combined immunodeficiency (SCID).

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 190-310, a region that may be conserved across isoforms; thus, the antibody could recognize multiple splice variants. Validate isoform specificity if needed.
  • For WB, a band near the predicted molecular weight of ~96 kDa is expected. Use appropriate loading controls and consider the expression levels in your target tissue or cell line.
  • In IHC-P, antigen retrieval conditions (e.g., citrate buffer pH 6.0 or EDTA buffer pH 9.0) should be optimized based on the tissue type. Nuclear staining pattern is anticipated.
  • For IF/ICC, permeabilization is required for nuclear access. Co-staining with a nuclear marker is recommended to confirm localization.
  • This antibody is validated for ELISA; consider its use in capture or detection formats after confirming binding in your specific assay setup.
