Biological Background: CTGF Function and Localization
- CCN family member 2, also known as connective tissue growth factor, hypertrophic chondrocyte-specific protein 24 (HCS24), and insulin-like growth factor-binding protein 8 (IGFBP-8).
- Major connective tissue mitoattractant secreted by vascular endothelial cells; promotes proliferation and differentiation of chondrocytes.
- Plays a critical role in osteogenesis by stimulating osteoblast differentiation (PubMed:39414788). Related references: PMID:39414788
- Mediates heparin- and divalent cation-dependent cell adhesion in various cell types including fibroblasts, myofibroblasts, endothelial, and epithelial cells.
- Enhances fibroblast growth factor-induced DNA synthesis.
- Localized to the extracellular space and secreted into the extracellular matrix.
- Expressed in bone marrow, thymic cells, and certain Wilms tumors; alternative splicing generates multiple isoforms.
- Glycoprotein with disulfide bonds; heparin-binding activity; associated with disease variants such as dwarfism.
Experimental Guidance and Technical Tips
- The immunogen (amino acids 100–200) is conserved across human, mouse, and rat CTGF, supporting the reported cross-reactivity; sequence alignment may be verified for additional species.
- For Western blot, detect a band at approximately 38 kDa in lysates from relevant tissues or cell lines; validate expression in the sample system of interest.
- In IHC-P, antigen retrieval and endogenous peroxidase blocking are recommended; test with positive control tissues (e.g., bone marrow) and optimize primary antibody dilution.
- For IF/ICC, fixation and permeabilization conditions should be optimized; a secreted target may require inhibition of protein secretion for cellular staining.
- This polyclonal antibody may be suitable for ELISA as either capture or detection reagent; pairwise testing with appropriate standards is advised.
