Biological Background: CTGF Function and Localization
- CTGF (Connective Tissue Growth Factor), also known as CCN2, is a member of the CCN family of matricellular proteins; alternative names include NOV2, HCS24, and IGFBP8 (IGF-binding protein 8).
- It functions as a major mitoattractant secreted by vascular endothelial cells, promoting proliferation and differentiation of chondrocytes and playing a critical role in osteogenesis. Related references: PMID:39414788
- Mediates heparin- and divalent cation-dependent cell adhesion in fibroblasts, myofibroblasts, endothelial, and epithelial cells.
- Enhances fibroblast growth factor-induced DNA synthesis.
- Localizes to the extracellular matrix and is secreted into the extracellular space.
- Expressed in bone marrow, thymic cells, and some Wilms tumors.
- Contains disulfide bonds and is a heparin-binding glycoprotein, reflecting its extracellular matrix association and role in connective tissue remodeling.
- Disease variants are associated with dwarfism.

- The immunogen spans amino acids 100–200 of human CTGF; the antibody may recognize both secreted and intracellular forms. For WB, a band of approximately 38 kDa is expected—validate in your specific sample matrix with appropriate loading controls.
- For immunofluorescence (IF/ICC, IF-P), consider permeabilization to detect intracellular CTGF; secreted protein may also be found associated with the cell surface or extracellular matrix.
- Given the secreted nature of CTGF, avoid cross-linking fixatives like formaldehyde for IHC/IF if extracellular staining is desired, as crosslinking may reduce accessibility.
- Cross-reactivity with rat has been reported; verify performance in your rat tissue/cell system by running appropriate controls.
