Biological Background: CST1 Function and Localization
- Cystatin-SN (CST1), also known as Cystatin-1 or salivary cystatin-SA-1, is a secreted cysteine protease inhibitor belonging to the cystatin superfamily.
- It plays a key role in regulating endogenous cysteine proteases, with stronger inhibition of papain and dipeptidyl peptidase I compared to the related cystatin S, while both inhibit ficin equally.
- The protein contains a signal peptide and two disulfide bonds characteristic of type 2 cystatins, contributing to its structural stability and function.
- Subcellular localization is predominantly extracellular; it is secreted into various body fluids.
- Tissue expression is specific: at the protein level, it is found in submandibular and sublingual saliva, tears, urine, and seminal fluid, but not in parotid saliva.
- The mature protein has a predicted molecular weight of approximately 16.4 kDa, migrating around 14 kDa under reducing SDS-PAGE conditions.
- Post-translational modifications include a signal peptide cleavage and disulfide bond formation, essential for its inhibitory activity.
Experimental Guidance and Technical Tips
- The immunogen used (amino acids 20–141) corresponds to the mature secreted protein; thus, the antibody is expected to detect the endogenous secreted form, but intracellular precursor detection may require specific treatment.
- For Western blot, due to the small predicted molecular weight (14 kDa), consider using a high-percentage acrylamide gel (e.g., 15%) and optimized low-molecular-weight transfer conditions (e.g., 0.2 µm PVDF membrane, shorter transfer times).
- In ELISA applications, validate the antibody pair with a recombinant CST1 standard and confirm specificity in complex biological matrices such as saliva or urine.
- Cross-reactivity has been reported only for human CST1; reactivity with mouse or other species is not predicted; thus, species-specific validation is necessary if used in non-human samples.
