Biological Background: CSRP3 Function and Localization

  • CSRP3, also known as Cardiac LIM protein (CLP) or Muscle LIM protein (MLP), is a member of the cysteine and glycine-rich protein family and contains two LIM zinc-binding domains, facilitating protein-protein interactions.
  • Acts as a positive regulator of myogenesis by serving as a cofactor for myogenic bHLH transcription factors (MYOD1, MYOG, MYF6), enhancing their DNA-binding and promoting muscle-specific gene expression.
  • In cardiomyocytes, plays a crucial structural role by organizing cytosolic components at the Z-line, acting as a scaffold that anchors calcineurin and enables mechanical stretch sensing and stress-activated calcineurin-NFAT signaling.
  • Binds directly to actin filaments, cross-linking them into bundles and protecting against cofilin-mediated depolymerization. Also bidirectionally modulates CFL2 activity, either enhancing or reducing actin depolymerization depending on the CSRP3:CFL2 ratio.

    PMID:24934443
  • In vitro inhibits the activity of protein kinase C alpha (PKC/PRKCA), suggesting a role in attenuating excessive cardiac stress signaling.

    PMID:27353086
  • An alternative isoform (isoform 2) is detected in striated muscle, where it may regulate early sarcomere organization and negatively influence myotube differentiation.
  • Subcellularly localizes to the nucleus, cytoplasm, and sarcomeric Z-lines, consistent with dual roles in transcriptional regulation and cytoskeletal organization.
  • Phosphorylation and metal-ion coordination are predicted post-translational modifications that likely modulate its activity and interactions.

Experimental Guidance and Technical Tips

  • The immunogen comprises nearly full-length human CSRP3 (aa 1-194), so the polyclonal antibody is expected to recognize multiple epitopes and may detect both isoforms.
  • For Western blotting, a predominant band around 21 kDa should appear in cardiac and skeletal muscle lysates from human, mouse, or rat; include appropriate positive and negative controls.
  • In immunohistochemistry (IHC-P), test standard antigen retrieval techniques (e.g., citrate buffer pH 6.0) and optimize primary antibody dilution; due to nuclear/cytoplasmic localization, expect staining in cardiomyocytes and slow-twitch myofibers.
  • For immunofluorescence/ICC, consider co-staining with sarcomeric markers (e.g., α-actinin) to verify Z-line localization; permeabilization method may influence detection.
  • ELISA applications may benefit from a blocking step tailored to the sample matrix; coated antigen or sandwich formats can be explored.
  • As with any polyclonal antibody, lot-to-lot validation is recommended, and species cross-reactivity should be confirmed in the specific experimental system.
