- CSRP3, also known as Cardiac LIM protein (CLP) or Muscle LIM protein (MLP), is a member of the cysteine and glycine-rich protein family and contains two LIM zinc-binding domains, facilitating protein-protein interactions.
- Acts as a positive regulator of myogenesis by serving as a cofactor for myogenic bHLH transcription factors (MYOD1, MYOG, MYF6), enhancing their DNA-binding and promoting muscle-specific gene expression.
- In cardiomyocytes, plays a crucial structural role by organizing cytosolic components at the Z-line, acting as a scaffold that anchors calcineurin and enables mechanical stretch sensing and stress-activated calcineurin-NFAT signaling.
- Binds directly to actin filaments, cross-linking them into bundles and protecting against cofilin-mediated depolymerization. Also bidirectionally modulates CFL2 activity, either enhancing or reducing actin depolymerization depending on the CSRP3:CFL2 ratio.
