Biological Background: CSK Function and Localization
- CSK is a non-receptor tyrosine-protein kinase that plays a critical role in regulating cell growth, differentiation, migration, and immune response.
- It phosphorylates the C-terminal tyrosine residues of Src-family kinases (SFKs) such as LCK, SRC, HCK, FYN, LYN, and YES1, promoting an inactive conformation through intramolecular interactions.
- CSK suppresses signaling from various surface receptors including T-cell receptor (TCR) and B-cell receptor (BCR) by maintaining SFKs in an inactive state.

- Subcellularly, CSK localizes to the cytoplasm and is recruited to the cell membrane via interactions with transmembrane or adapter proteins for SFK inhibition.
- Tissue expression is detected in lung and macrophages, consistent with its role in immune regulation.
- The protein contains SH2 and SH3 domains and requires ATP, magnesium, or manganese for kinase activity; it is subject to acetylation and phosphorylation modifications.

- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval and permeabilization protocols may need optimization; cytoplasmic and membrane staining patterns are anticipated.
