  • CSK is a non-receptor tyrosine-protein kinase that plays a critical role in regulating cell growth, differentiation, migration, and immune response.
  • It phosphorylates the C-terminal tyrosine residues of Src-family kinases (SFKs) such as LCK, SRC, HCK, FYN, LYN, and YES1, promoting an inactive conformation through intramolecular interactions.
  • CSK suppresses signaling from various surface receptors including T-cell receptor (TCR) and B-cell receptor (BCR) by maintaining SFKs in an inactive state.

  • Subcellularly, CSK localizes to the cytoplasm and is recruited to the cell membrane via interactions with transmembrane or adapter proteins for SFK inhibition.
