Biological Background: CSF1 Function and Localization
- CSF1, also known as macrophage colony-stimulating factor (M-CSF) or Lanimostim, is a cytokine and growth factor that exists in both secreted and proteoglycan membrane-bound forms (PG-M-CSF).
- It is essential for the survival, proliferation, and differentiation of hematopoietic precursor cells, particularly promoting the formation of mononuclear phagocytes such as macrophages and monocytes.
- CSF1 plays a key role in innate immunity and inflammation by stimulating the release of pro-inflammatory chemokines.
- It is critical for bone homeostasis, regulating osteoclast proliferation, differentiation, and bone resorption, and is required for normal bone development.
- CSF1 is necessary for normal male and female fertility.
- The cytokine influences cell morphology and motility by promoting reorganization of the actin cytoskeleton, formation of membrane ruffles, and cell adhesion and migration.
- It also participates in lipoprotein clearance.
- Subcellularly, CSF1 is found at the cell membrane, secreted into the extracellular space, and undergoes proteolytic processing. It is heavily glycosylated and phosphorylated, contributing to its observed molecular weight of approximately 60 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 33-190 of human CSF1, covering a portion of the mature secreted protein. This design may help in capturing both the processed and full-length forms.
- For Western blotting, consider using conditioned medium or cell lysates from human, mouse, or rat samples. Due to glycosylation, the detected band may appear larger than the calculated molecular weight.
- For ELISA applications, validate the detection range using a recombinant CSF1 standard curve. The antibody’s polyclonal nature may provide robust signal for capture or detection formats.
- Since cross-reactivity with mouse and rat is noted, verify reactivity in relevant sample systems if species-specific detection is critical.
