Biological Background: CRYM Function and Localization
- CRYM encodes ketimine reductase mu-crystallin, also known as NADP-regulated thyroid-hormone-binding protein (THBP).
- It functions as an NAD(P)H-dependent oxidoreductase that reduces cyclic ketimine substrates, including delta(1)-piperideine-2-carboxylate (P2C) and delta(1)-pyrroline-2-carboxylate (Pyr2C), indicating a central role in lysine and glutamate metabolism. Related references: PMID:21332720 PMID:25931162
- Additional substrates include delta(2)-thiazoline-2-carboxylate (T2C), 3,4-dehydrothiomorpholine-3-carboxylate (AECK), and (R)-lanthionine ketimine (LK), with LK reduced at a lower rate. Related references: PMID:25931162
- Also catalyzes the reduction of (S)-cystathionine ketimine (CysK) (by similarity).
- The protein is primarily localized in the cytoplasm.
- Expressed in neural tissues, muscle, kidney, and the inner ear; mutations in CRYM are associated with non-syndromic deafness (DFNA40). Related references: PMID:1384048 PMID:12471561
- Keywords include alternative names (THBP), oxidoreductase activity, NAD/NADP binding, and disease variant connection.
Experimental Guidance and Technical Tips
- As a polyclonal antibody raised against nearly full-length recombinant CRYM protein, it is expected to recognize multiple epitopes, providing robust detection in Western blot and ELISA. Consider optimizing antibody dilution for your specific sample type and assay conditions.
- The predicted molecular weight is approximately 34 kDa; verify the band size in your experimental system, and note potential post-translational modifications or isoform variations.
- For ELISA applications, use as capture or detection antibody after validating specificity in your target matrix. Titrate antibody concentrations to achieve optimal signal-to-noise ratios.
