Biological Background: CRYGC Function and Localization
- CRYGC, also known as gamma-crystallin C, gamma-C-crystallin, gamma-crystallin 2-1, and gamma-crystallin 3, is a member of the beta/gamma-crystallin family.
- It functions as a dominant structural protein of the vertebrate eye lens, contributing to lens transparency and refractive index.
- Crystallins are encoded by a gene family that includes several gamma-crystallin genes (CRYGA, CRYGB, etc.), which share high sequence similarity and lens-specific expression.
- Mutations in CRYGC and other crystallins are associated with congenital cataracts, as indicated by the UniProt keywords "Cataract" and "Disease variant."
- The protein has a molecular weight of approximately 21 kDa (calculated from sequence) and is subject to post-translational modifications such as methylation.
- CRYGC is exclusively expressed in the lens, consistent with its role in maintaining lens structure and function.
- It is identified in proteomics studies and forms part of the reference proteome for human.
Experimental Guidance and Technical Tips
- This polyclonal antibody was raised against a recombinant fusion protein containing amino acids 1-174 of human CRYGC. For Western blot, a typical starting dilution of 1:500-1:2000 may be evaluated with lens tissue lysates or cell lines overexpressing CRYGC.
- In immunofluorescence/immunocytochemistry, consider using a dilution of 1:50-1:200 and optimize fixation and permeabilization conditions for lens cells or tissue sections.
- For ELISA, the antibody can be tested as a capture or detection reagent; validate with recombinant CRYGC protein or lens extracts.
- Cross-reactivity with mouse and rat is reported; however, sequence homology should be checked for specific applications, and validation in the relevant species is recommended.
