Biological Background: Alpha-crystallin B chain Function and Localization
- Alpha-crystallin B chain (CRYAB), also known as HspB5, is a member of the small heat shock protein family. It is also referred to as renal carcinoma antigen NY-REN-27 and Rosenthal fiber component.
- Functions as a molecular chaperone, preventing the aggregation of various proteins under stress conditions. It may contribute to the transparency and refractive index of the lens.
- Belongs to the heat shock protein family B (small HSP) and is specifically classified as HSPB5.
- Subcellularly localizes to the cytoplasm, nucleus, secreted, and lysosome compartments, indicating diverse functional roles.
- Expressed in the lens as well as other tissues (PubMed:2387586, PubMed:838078). Also expressed in myocardial tissue (PubMed:28493373). Related references: PMID:2387586 PMID:838078 PMID:28493373
- Undergoes various post-translational modifications including phosphorylation, acetylation, and oxidation. It is associated with several human diseases such as cardiomyopathy, cataract, and myofibrillar myopathy.
Experimental Guidance and Technical Tips
- For Western blotting, consider using a dilution range of 1:500-1:2000, with further optimization based on target abundance and sample type.
- Validate cross-reactivity in your specific mouse or rat tissue samples, as reactivity may vary with tissue preparation and expression levels.
- For ELISA, titrate the antibody to determine the optimal coating concentration suitable for your assay format.
- As a polyclonal antibody, verify lot-to-lot consistency for critical quantitative assays.
