Biological Background: CRY1 Function and Localization
- CRY1 (Cryptochrome-1) is a transcriptional repressor and a core component of the circadian clock, forming the negative limb of the transcription/translation feedback loop (TTFL). It heterodimerizes with PER proteins to interact with and inhibit the CLOCK/BMAL1 complex.
- The circadian clock governs ~24-hour rhythms in gene expression, regulating metabolism, sleep, body temperature, and many physiological processes. CRY1, along with CRY2, PER1-3, and CLOCK/BMAL1, drives this molecular oscillator.
- CRY1 and CRY2 have redundant yet distinct functions: CRY1 is a more potent transcriptional repressor in cerebellum and liver, while CRY2 tunes the period of the SCN oscillator. CRY1 is essential for intercellular network development and rhythm synchrony.
- Subcellular localization: CRY1 is found in both the cytoplasm and nucleus. It can translocate clock proteins (e.g., PER) to the nucleus and act as a gatekeeper for CLOCK-BMAL1 at DNA-binding sites.
- Post-translational modifications: CRY1 undergoes phosphorylation and is a chromophore-binding flavoprotein (FAD). It is also a photoreceptor protein involved in light entrainment of the circadian clock.
- Beyond the clock, CRY1 represses glucocorticoid receptor NR3C1/GR transcriptional activity and plays a role in glucose and lipid metabolism. In liver, it mediates circadian regulation of cAMP signaling and gluconeogenesis, inhibiting FOXO1 and CREB1 pathways.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal fragment (aa 496-586) of human CRY1; this region may be less conserved between CRY1 and CRY2, potentially offering isoform specificity. Validate in relevant sample systems (e.g., SCN-containing brain extracts, liver lysates) with positive and negative controls.
- This antibody is suitable for Western blotting (WB), immunohistochemistry-paraffin (IHC-P), and ELISA. For WB, a band near 66 kDa is expected; consider loading controls such as GAPDH or β-actin. For IHC-P, appropriate antigen retrieval and permeabilization steps may be required due to nuclear localization.
