Biological Background: CRTC2 Function and Localization

  • CRTC2 (CREB-regulated transcription coactivator 2), also known as TORC2 or TORC-2, is a ~73 kDa transcriptional coactivator belonging to the CRTC family.
  • It acts as a potent coactivator for CREB1, enhancing transcription through both consensus and variant cAMP response element (CRE) sites, and functions independently of CREB1 Ser-133 phosphorylation in the SIK/TORC signaling pathway.
  • CRTC2 enhances the interaction between CREB1 and TAF4, and also serves as a coactivator for TAX-mediated activation of human T-cell leukemia virus type 1 (HTLV-1) long terminal repeats (LTR).
  • As a key component of the LKB1/AMPK/TORC2 signaling pathway, it regulates gluconeogenesis and promotes mitochondrial biogenesis by acting as a coactivator of PPARGC1A in muscle cells.
  • Subcellular localization is dynamic; CRTC2 shuttles between the cytoplasm and nucleus, depending on phosphorylation status.
  • Post-translational modifications include acetylation, phosphorylation, methylation, and ubiquitin-like conjugation (ISGylation), which modulate its activity and stability.
  • Tissue distribution is broad, with highest expression in thymus, muscle, and insulinomas; also found in B and T lymphocytes, spleen, ovary, lung, and at lower levels in brain, colon, heart, kidney, prostate, small intestine, stomach, and liver.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 16–126, an N-terminal region; verify antibody specificity using positive (e.g., thymus, muscle lysates) and negative controls.
  • For Western blot, a clear band at approximately 73 kDa is expected; use reducing SDS-PAGE conditions and load appropriate protein amounts.
  • In ELISA applications, consider both direct and sandwich formats; perform titration experiments to determine optimal antibody dilution for your specific assay conditions.
  • CRTC2 expression is inducible and cell-type dependent; include relevant stimuli (e.g., cAMP-elevating agents) if activation-dependent detection is required.
