- CRTC2 (CREB-regulated transcription coactivator 2), also known as TORC2 or TORC-2, is a ~73 kDa transcriptional coactivator belonging to the CRTC family.
- It acts as a potent coactivator for CREB1, enhancing transcription through both consensus and variant cAMP response element (CRE) sites, and functions independently of CREB1 Ser-133 phosphorylation in the SIK/TORC signaling pathway.
- CRTC2 enhances the interaction between CREB1 and TAF4, and also serves as a coactivator for TAX-mediated activation of human T-cell leukemia virus type 1 (HTLV-1) long terminal repeats (LTR).
- As a key component of the LKB1/AMPK/TORC2 signaling pathway, it regulates gluconeogenesis and promotes mitochondrial biogenesis by acting as a coactivator of PPARGC1A in muscle cells.
- Subcellular localization is dynamic; CRTC2 shuttles between the cytoplasm and nucleus, depending on phosphorylation status.
- Post-translational modifications include acetylation, phosphorylation, methylation, and ubiquitin-like conjugation (ISGylation), which modulate its activity and stability.
