- CRTC1 (CREB-regulated transcription coactivator 1), also known as MECT1 or TORC1, is a transcriptional coactivator that enhances CREB1-mediated transcription at cAMP response element (CRE) sites, functioning in the SIK/TORC signaling pathway.
- It acts independently of CREB1 Ser-133 phosphorylation, enhancing CREB1 interaction with TAF4, and regulates specific genes like StAR and PGC1alpha, thereby inducing mitochondrial biogenesis.
- In the hippocampus, CRTC1 is required for late-phase long-term potentiation (L-LTP) maintenance at Schaffer collateral-CA1 synapses and may support dendritic growth in developing cortical neurons (By similarity).
- Together with SIK1, it regulates light-induced entrainment of the circadian clock by coactivating CREB-mediated transcription of PER1.
- CRTC1 serves as a coactivator for the HTLV-1 Tax protein, enhancing viral LTR transcription during microbial infection.
- The protein localizes to both cytoplasm and nucleus and is subject to alternative splicing and phosphorylation; it is highly expressed in adult and fetal brain, especially prefrontal cortex and cerebellum, with very low expression elsewhere.
