Biological Background: Cornulin Function and Localization
- Cornulin (CRNN) is a 53 kDa putative calcium-binding protein, also known as SEP53, DRC1, or PDRC1, belonging to the fused-type S100 protein family.
- It promotes cell proliferation and G1/S cell cycle progression, and induces the expression of the cell cycle regulator CCND1 (cyclin D1) through activation of NFKB1 and PI3K/AKT signaling pathways. Related references: PMID:30009832
- CRNN is specifically expressed in squamous epithelia, including the granular layer of the epidermis, esophagus, foreskin, scalp skin, inner root sheath of hair follicles, and primary keratinocytes. Related references: PMID:30009832
- The protein localizes to the cytoplasm and contains calcium-binding EF-hand motifs, suggesting roles in calcium-dependent signaling or structural integrity of squamous epithelia.
- Its expression is significantly downregulated or absent in esophageal and oral squamous cell carcinomas, as well as in laryngopharyngeal reflux conditions, indicating a potential tumor-suppressive or differentiation marker role. Related references: PMID:30009832
- The primary isoform has a theoretical molecular weight of approximately 53.5 kDa, consistent with the observed 54 kDa in Western blot analysis.
Experimental Guidance and Technical Tips
- For Western blot, the predicted band is ~54 kDa. Ensure proper protein extraction from squamous epithelial cell lines or tissues expressing CRNN, such as primary keratinocytes or esophagus tissue.
- For ELISA, consider using recombinant CRNN protein as a standard for quantification and to establish assay specificity.
- Validate antibody performance in the relevant sample system by including positive controls (e.g., skin or esophageal tissue lysate) and negative controls (e.g., lung or heart tissue with low expression).
- Since CRNN expression is lost in many carcinomas, this antibody may be useful for differentiating normal versus malignant squamous epithelial tissues in research settings.
