Biological Background: CRMP2/DPYSL2 Function and Localization
- Dihydropyrimidinase-related protein 2 (CRMP2/DPYSL2), also known as collapsin response mediator protein 2, N2A3, unc-33-like phosphoprotein 2 (ULIP-2), is encoded by the DPYSL2 gene in humans.
- Plays a central role in neuronal development and polarity, axon growth and guidance, growth cone collapse, and cell migration. It is necessary for signaling by class 3 semaphorins and subsequent cytoskeletal remodeling.
- May also participate in endocytosis.
- Subcellular localization includes cytosol, cytoskeleton, and membrane.
- Ubiquitously expressed across tissues.
- Subject to multiple post-translational modifications; annotated as a phosphoprotein, methylated, and S-nitrosylated. Involved in neurogenesis and differentiation, with alternative splicing generating isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 473–572 of human CRMP2/DPYSL2; consider using full-length protein or the relevant peptide for competitive assays if needed.
- For WB, a band at ~62 kDa is expected; validate in neuronal cell lines or tissues from human, mouse, or rat.
