Biological Background: CRM1/XPO1 Function and Localization
- Exportin-1 (CRM1/XPO1) is a member of the importin-beta superfamily of nuclear transport receptors, encoded by the XPO1 gene (GeneID: 7514). It is also known as chromosome region maintenance 1 protein homolog.
- CRM1 mediates the nuclear export of proteins containing a leucine-rich nuclear export signal (NES) and RNAs. In the nucleus, it binds cooperatively to the NES cargo and Ran-GTP, then docks at the nuclear pore complex via nucleoporins.
- Upon translocation to the cytoplasm, the complex disassembles: Ran-GTP is hydrolyzed to Ran-GDP by RanGAP1 and RanBP1, triggering cargo release. The asymmetric distribution of Ran-GTP/GDP drives export directionality.
- CRM1 is involved in U3 snoRNA transport from Cajal bodies to nucleoli, binding to late precursor U3 snoRNA with a TMG cap.
- During viral infection, CRM1 exports unspliced or incompletely spliced RNAs of HIV-1, HTLV-1, and influenza A. It interacts with HIV-1 Rev and HTLV-1 Rex proteins and facilitates Rex multimerization.
- Subcellularly, CRM1 localizes to the cytoplasm, nucleoplasm, Cajal bodies, and nucleolus.
- It is widely expressed in tissues including heart, brain, placenta, lung, liver, skeletal muscle, pancreas, spleen, thymus, prostate, testis, ovary, small intestine, colon, and peripheral blood leukocytes, but is not expressed in the kidney.
- Key post-translational modifications include phosphorylation and acetylation (as indicated by keywords); the protein is also involved in host-virus interactions, mRNA transport, and protein transport.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 972–1071 of human CRM1/XPO1 (C-terminal region). When using this antibody in Western blotting, a band of approximately 123 kDa is expected; consider using a 6–10% SDS-PAGE gel for optimal resolution.
- For immunofluorescence/ICC, CRM1 exhibits both cytoplasmic and nuclear (including nucleolar and Cajal body) staining; fixation and permeabilization conditions should be optimized to preserve these compartments.
- The antibody is validated for WB, IF/ICC, and ELISA and cross-reacts with mouse and rat; it can be considered for studies of nuclear transport, viral-host interactions, and cell cycle regulation in these species.
- Since the immunogen is a synthetic peptide, it is advisable to confirm specificity using positive and negative control lysates or knockdown/knockout validation in the relevant sample system.
