Biological Background: Corticotropin-Releasing Hormone (CRH) Function and Localization
- CRH, also known as corticotropin-releasing factor (CRF) or corticoliberin, is a secreted peptide hormone primarily produced by the hypothalamus and placenta.
- It plays a central role in whole-body adaptation to stress by binding to CRHR1 and, to a lesser extent, CRHR2. This triggers the neuroendocrine, autonomic, and behavioral stress responses, including activation of the hypothalamic-pituitary-adrenal (HPA) axis and subsequent adrenocorticotropic hormone (ACTH) release. - For Western blot, the predicted molecular weight is approximately 21 kDa; a band near this size in hypothalamus or placenta lysates can be expected. Inclusion of protease inhibitors is recommended due to rapid hormone processing.
- In ELISA, titrate the antibody under your specific assay conditions and validate against known CRH standards. Cross-reactivity with mouse and rat CRH allows use in rodent stress models.
